The release of the chemical, called butyl acetate, occurred after the crash ignited a column of black smoke, later confirmed by Kentucky State Police to be a toxic cloud.
A portion of the Kentucky River also caught fire from the chemical spill.
Police say emergency crews are working to extinguish the flames, and people closest to the crash have been evacuated. Others in the area have been told to stay in their homes with the windows closed, and to stuff towels under their doors.
No injuries have been reported.
Both the parked locomotive and the runaway cars were unmanned.
